Custom DNS Server to PFSense

Hey I'm using unbound on my PFsense with PiHole and I'm just wondering if I need to set the port or is the LAN interface of my PFsense enough?

Depends on what port unbound is listening on. If it's the standard 53 then no port is needed, if not then add the port.

You can add 53 without hurting anything if unbound is listening on 53.
