Not without some advanced configuration. That's why we put that large warning in the file.

so i'm in a big trouble if i can't have multiple web site easily...
i have replace the original lighttpd.conf file and i think the pihole is working
after a reboot, it's back to not working like before

su -s /bin/sh -s "cat /etc/os-release" "pi"
Work if you use the password for pi? It may be a segmentation fault in another binary.

Okay, then the problem is with the underlying operating system and not with Pi-hole itself.
Looks like you'll need to reinstall the OS if su or /bin/sh is broken.
Edlt: It's su that is broken. the /bin/sh was never even called.
so i've reinstall almost everything on my RPI and it's look like to work. now i have to figure out how to make it work with several websites.
