Hello, I have two pi that are used at home, I was thinking maybe replacing them with a NUC, do you know if using pi-hole on a regular HDD can slow down?
And using openvpn client on the same machine can be a problem?

It should not. Pi-hole run completely in memory and only access the storage on a regular base when it saves data to the long-term database. Normally this happens once a minute. But DNS resolution should not be affected during that time.
It should not. But clearly it depends on the size of your network and the amount of traffic that will be routed via the device. But for normal home network you should not see any issues.
